Understanding Delta-6 Desaturase Activity: Normal vs Optimal Ranges

Also known as: delta-6-desaturase activity

MetabolicUnit: ratio

?What is Delta-6 Desaturase Activity?

Delta-6 desaturase (D6D) is a rate-limiting enzyme in polyunsaturated fatty acid metabolism, converting linoleic acid (LA) to gamma-linolenic acid (GLA) and alpha-linolenic acid (ALA) to stearidonic acid — the first steps in omega-6 and omega-3 elongation pathways respectively. D6D activity is inferred from fatty acid ratios in blood (e.g., GLA/LA or AA/DGLA ratios) rather than measured directly.

!Why It Matters

Impaired D6D activity limits the body's ability to produce long-chain omega-3s (EPA, DHA) from plant-based ALA, and beneficial GLA from LA. D6D activity is reduced by insulin resistance, zinc deficiency, excess trans fats, magnesium deficiency, and ageing. Measuring D6D activity via fatty acid profiling helps identify why some individuals have poor omega-3 status despite adequate dietary intake.

Range TypeMinMaxUnitNote

Lab normal ranges may vary between laboratories. Optimal and longevity targets are based on research literature and should be interpreted with your physician.

Symptoms of Imbalance

  • Poor conversion of plant-based omega-3s (ALA) to EPA/DHA
  • Dry skin, eczema, or inflammatory skin conditions
  • Premenstrual syndrome — GLA deficiency-related
  • Nerve pain and dry eyes

How to Improve Your Levels

  • 1Consume pre-formed EPA/DHA (oily fish, algae oil) rather than relying on ALA conversion
  • 2Supplement with GLA (evening primrose oil or borage oil) to bypass D6D limitation
  • 3Address zinc and magnesium deficiency — both are D6D cofactors
  • 4Eliminate trans fats — they competitively inhibit D6D
  • 5Improve insulin sensitivity — insulin resistance directly impairs D6D activity

When to Test

Poor omega-3 status despite supplementation; inflammatory conditions not responding to omega-3s; fatty acid profile evaluation; suspected enzyme-level fatty acid metabolism impairment.
