?What is Nicotinamide?
Nicotinamide (niacinamide, vitamin B3 amide form) is one of three main forms of vitamin B3, alongside nicotinic acid and nicotinamide riboside. It is the primary precursor for NAD+ (nicotinamide adenine dinucleotide) synthesis via the salvage pathway. Unlike nicotinic acid, nicotinamide does not cause flushing and is widely used dermatologically (50 mg topical) and for NAD+ precursor supplementation. It also has anti-inflammatory properties via PARP-1 inhibition.

NAD+ is essential for cellular energy production (mitochondrial OXPHOS), DNA repair (PARP-1 and SIRT1 activity), and signalling. NAD+ levels decline with age by ~50% by age 60, contributing to mitochondrial dysfunction, impaired DNA repair, and metabolic decline. Nicotinamide supports NAD+ replenishment and is being studied for skin cancer prevention, Alzheimer's disease, and metabolic ageing.
